Polyamine Metabolism Is Involved in the Direct Regeneration of Shoots from Arabidopsis Lateral Root Primordia

چکیده
Plants can be regenerated from various explants/tissues via de novo shoot meristem formation. Most of these regeneration pathways are indirect and involve callus Besides plant hormones, the role polyamines (PAs) has been implicated in processes. Interestingly, lateral root primordia (LRPs) Arabidopsis directly converted to meristems by exogenous cytokinin application. In this system, no formation takes place. We report that level PAs, especially spermidine (Spd), increased during conversion application Spd improved its efficiency. The high endogenous could due enhanced synthesis as indicated augmented relative expression PA genes (AtADC1,2, AtSAMDC2,4, AtSPDS1,2) process. However, effect PAs on might also dependent their catabolism. POLYAMINE OXIDASE 5 (AtPAO5) was shown specifically process ectopic overexpression LRP-to-shoot This correlated with accumulation roots ROS converting LRPs. potential ways how PAO5 may influence direct organogenesis LRPs discussed.

Quiescent center initiation in the Arabidopsis lateral root primordia is dependent on the SCARECROW transcription factor.
Lateral root formation is an important determinant of root system architecture. In Arabidopsis, lateral roots originate from pericycle cells, which undergo a program of morphogenesis to generate a new lateral root meristem. Despite its importance for root meristem organization, the onset of quiescent center (QC) formation during lateral root morphogenesis remains unclear. Here, we used live 3D ...
متن کاملLRP7, a Gene Expressed in Lateral and Adventitious Root Primordia of Arabidopsis
We describe a gene that is expressed in lateral and adventitious root primordia of Arabidopsis. The gene was identified by expression of a transposon-borne promoterless P-glucuronidase gene in lateral root primordia. The gene, designated LRP7 for lateral root primordium 1, and its corresponding cDNA were cloned and sequenced. The expression pattern of the gene in lateral root primordia was conf...

We describe a gene that is expressed in lateral and adventitious root primordia of Arabidopsis. The gene was identified by expression of a transposon-borne promoterless beta-glucuronidase gene in lateral root primordia. The gene, designated LRP1 for lateral root primordium 1, and its corresponding cDNA were cloned and sequenced. The expression pattern of the gene in lateral root primordia was c...
متن کاملHeterologous expression and biochemical characterization of a polyamine oxidase from Arabidopsis involved in polyamine back conversion.
Polyamine oxidase (PAO) is a flavin adenine dinucleotide-dependent enzyme involved in polyamine catabolism. Animal PAOs oxidize spermine (Spm), spermidine (Spd), and/or their acetyl derivatives to produce H2O2, an aminoaldehyde, and Spd or putrescine, respectively, thus being involved in a polyamine back-conversion pathway. On the contrary, plant PAOs that have been characterized to date oxidiz...
متن کاملLateral root formation and growth of Arabidopsis is redundantly regulated by cytokinin metabolism and signalling genes
The plant root system is important for the uptake of water and nutrients and the anchoring of plants in the soil. Lateral roots (LRs) contribute considerably to root system architecture. Their post-embryonic formation is regulated by hormones and environmental cues. The hormone cytokinin influences LR formation and growth in Arabidopsis thaliana on different levels by disturbing cell division a...
